I've seen foam blocks used to block the register boots for a duct leakage test. Anyone know where to get these?

You can buy 1" thick sheets of neoprene for rounds, 2" thick for rectangular.   We made rounds, 2x10, 2x12, 4x10, 4x12, 6x10's, etc.   Sometimes you run into an odd size (8x10) which can be sealed using two 4x10's.   It adds a lot of equipment to your vehicle but saves a lot of time.

Similarly, I contacted a local mattress making/repair place and I had them cut to size - very inexpensive, I thought.

I then use sandwich bags (free-of course) to stuff them into - otherwise you might lose about 6 cfm through each. They last quite a while if not abused.

I found cheaper blocks at several upholstery shops in different areas of the country. You may need to search for smaller shops. I bought large trash bags of scrap foam for $5 each. Some of the scrap wasn't usable, some needed to be trimmed. For $15 I got thirty or more blocks. I use them inside small plastic trash bags folded over. The seal is very good.

Aero Seal sells quality foam panels you can cut into any size and re-use them for testing.
